Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_170707.4(LMNA):c.369G>T (p.Lys123Asn), citing Ambry Variant Classification Scheme 2023: The p.K123N variant (also known as c.369G>T), located in coding exon 2 of the LMNA gene, results from a G to T substitution at nucleotide position 369. The lysine at codon 123 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
