This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 737, where C is replaced by G; at the protein level this means replaces proline at residue 246 with arginine — a missense variant. Submitter rationale: The p.P246R variant (also known as c.737C>G), located in coding exon 7 of the PTEN gene, results from a C to G substitution at nucleotide position 737. The proline at codon 246 is replaced by arginine, an amino acid with dissimilar properties. This variant demonstrated low intracellular protein abundance in a massively parallel functional assay (Matreyek KA et al. Nat Genet, 2018 Jun;50:874-882). In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955).
