This variant lies in the APC gene (transcript NM_000038.6) at coding-DNA position 1231, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 411 with lysine — a missense variant. Submitter rationale: The p.E411K variant (also known as c.1231G>A), located in coding exon 9 of the APC gene, results from a G to A substitution at nucleotide position 1231. The glutamic acid at codon 411 is replaced by lysine, an amino acid with similar properties. This variant was observed in an individual with early onset-breast cancer amongst a cohort of 1781 non-Ashkenazi Jewish individuals undergoing BRCA1/2 gene testing based on a personal history of breast cancer (Tung N et al. Cancer, 2015 Jan;121:25-33). This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
