NM_000057.4(BLM):c.3505G>T (p.Ala1169Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A1169S variant (also known as c.3505G>T), located in coding exon 17 of the BLM gene, results from a G to T substitution at nucleotide position 3505. The alanine at codon 1169 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
