Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.76G>C (p.Gly26Arg), citing Ambry Variant Classification Scheme 2023: The p.G26R variant (also known as c.76G>C), located in coding exon 1 of the ALK gene, results from a G to C substitution at nucleotide position 76. The glycine at codon 26 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
