Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3172C>T (p.Leu1058Phe), citing Ambry Variant Classification Scheme 2023: The p.L1058F variant (also known as c.3172C>T), located in coding exon 19 of the DICER1 gene, results from a C to T substitution at nucleotide position 3172. The leucine at codon 1058 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
