NM_177438.3(DICER1):c.5621dup (p.Tyr1874Ter) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.5621dupA variant, located in coding exon 26 of the DICER1 gene, results from a duplication of A at nucleotide position 5621, causing a translational frameshift with a predicted alternate stop codon (p.Y1874*). This alteration occurs at the 3' terminus of theDICER1 gene, is not expected to trigger nonsense-mediated mRNAdecay, and only impacts the last 49 amino acids of the protein. The exact functional effect of this alteration is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
