Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.1997A>T (p.Lys666Ile), citing Ambry Variant Classification Scheme 2023: The p.K666I variant (also known as c.1997A>T), located in coding exon 11 of the PMS2 gene, results from an A to T substitution at nucleotide position 1997. The lysine at codon 666 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
