NM_000535.7(PMS2):c.960T>A (p.Phe320Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F320L variant (also known as c.960T>A), located in coding exon 9 of the PMS2 gene, results from a T to A substitution at nucleotide position 960. The phenylalanine at codon 320 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
