Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.3644A>G (p.His1215Arg), citing Ambry Variant Classification Scheme 2023: The p.H1215R variant (also known as c.3644A>G), located in coding exon 22 of the PTCH1 gene, results from an A to G substitution at nucleotide position 3644. The histidine at codon 1215 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
