NM_001369.3(DNAH5):c.13570G>A (p.Asp4524Asn) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D4524N variant (also known as c.13570G>A), located in coding exon 78 of the DNAH5 gene, results from a G to A substitution at nucleotide position 13570. The aspartic acid at codon 4524 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
