Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.3181C>G (p.His1061Asp), citing Ambry Variant Classification Scheme 2023: The p.H1061D variant (also known as c.3181C>G), located in coding exon 25 of the BUB1 gene, results from a C to G substitution at nucleotide position 3181. The histidine at codon 1061 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
