NM_001232.4(CASQ2):c.776A>G (p.Glu259Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E259G variant (also known as c.776A>G), located in coding exon 7 of the CASQ2 gene, results from an A to G substitution at nucleotide position 776. The glutamic acid at codon 259 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
