NM_014297.5(ETHE1):c.461del (p.Asp154fs) was classified as Likely pathogenic for Metabolic acidosis; Rest dyspnea; Cardiomyopathy; Ethylmalonic encephalopathy by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the ETHE1 gene (transcript NM_014297.5) at coding-DNA position 461, deleting one base; at the protein level this means shifts the reading frame starting at aspartic acid residue 154, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The frameshift variant c.461del (p.Asp154ValfsTer4) in ETHE1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Asp154ValfsTer4 variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. This variant causes a frameshift starting with codon Aspartic Acid 154, changes this amino acid to Valine residue, and creates a premature Stop codon at position 4 of the new reading frame, denoted p.Asp154ValfsTer4. Loss of function variants have been previously reported to be disease causing. For these reasons, this variant has been classified as Likely Pathogenic.
