NM_006493.4(CLN5):c.75_81del (p.Trp26fs) was classified as Likely pathogenic for Frequent falls; Proptosis; Neuronal ceroid lipofuscinosis 5 by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The frameshift deletion p.W26Gfs*86 in CLN5 (NM_006493.4) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.W26Gfs*86 variant is novel (not in any individuals) in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing. This variant causes a frameshift starting with codon Tryptophan 26, changes this amino acid to Glycine residue, and creates a premature Stop codon at position 86 of the new reading frame, denoted p.Trp26GlyfsTer86. For these reasons, this variant has been classified as Likely Pathogenic.
