Likely pathogenic for Abnormal blistering of the skin; Epidermolysis bullosa simplex with nail dystrophy — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_201384.3(PLEC):c.1264-1G>A, citing ACMG Guidelines, 2015: The missense variant in c.11261G>A (p.Arg3754His) in PLEC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Arg3754His variant is reported with the allele frequency (0.005%) in the gnomad and novel in 1000 genome database. The amino acid Arg at position 3754 is changed to a His changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted to be damaging by both SIFT and PolyPhen2. The residue is conserved across species. The amino acid change p.Arg3754His in PLEC is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Variant of Uncertain Significance.
