This variant lies in the STAT1 gene (transcript NM_007315.4) at coding-DNA position 500, where A is replaced by C; at the protein level this means replaces glutamine at residue 167 with proline — a missense variant. Submitter rationale: The missense variant c.500A>C (p.Gln167Pro) in STAT1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Gln167Pro variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. The amino acid Gln at position 167 is changed to a Pro changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted to be damaging by both SIFT and PolyPhen2. The residue is conserved across species. The amino acid change p.Gln167Pro in STAT1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Variant of Uncertain Significance (VUS).
