Uncertain significance for Breast carcinoma; Breast-ovarian cancer, familial, susceptibility to, 1 — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_024675.4(PALB2):c.3137T>C (p.Leu1046Pro), citing ACMG Guidelines, 2015: The missense variant in c.3137T>C(p.Leu1046Pro) in PALB2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The amino acid Leu at position 1046 is changed to a Pro changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted to be damaging by both SIFT and PolyPhen2. The residue is conserved across species. The amino acid change p.Leu1046Pro in PALB2 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The p.Leu1046Pro variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. For these reasons, this variant has been classified as Uncertain Significance
