Likely pathogenic for Abnormality of the kidney; Nephropathic cystinosis — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_004937.3(CTNS):c.120dup (p.Asn41fs), citing ACMG Guidelines, 2015: The observed frameshift c.120dupp.Asn41GlnfsTer19 variant in CTNS gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is absent in gnomAD Exomes. This variant causes a frameshift starting with codon Asparagine 41, changes this amino acid to Glutamine residue, and creates a premature Stop codon at position 19 of the new reading frame, denoted p.Asn41GlnfsTer19. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing. For these reasons, this variant has been classified as Likely Pathogenic.
