Likely pathogenic for Interstitial lung disease due to ABCA3 deficiency — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001089.3(ABCA3):c.2741A>G (p.Lys914Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCA3 c.2741A>G (p.Lys914Arg) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 192470 control chromosomes. c.2741A>G has been reported in the literature in homozygous and compound heterozygous individuals affected with Pulmonary surfactant metabolism dysfunction (e.g. Hamvas_2009, Kitazawa_2013, Wambach_2014, Kumata_2022). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 19647838, 23443156, 24871971, 36376106). No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
