Submitter rationale: Variant summary: POU1F1 c.3G>C (p.Met1Ile) alters the initiation codon and is predicted to result either in absence of the protein or truncation of the encoded protein due to translation initiation at a downstream codon. The next downstream in-frame initiation codon is at Met 27. Activation of the potential downstream translation initiation site would result in a shortened protein missing the first 26 amino acids. Two of three in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250646 control chromosomes.
