Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.1077T>G (p.Phe359Leu), citing Ambry Variant Classification Scheme 2023: The p.F359L variant (also known as c.1077T>G), located in coding exon 12 of the RTEL1 gene, results from a T to G substitution at nucleotide position 1077. The phenylalanine at codon 359 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
