Uncertain significance — the classification assigned by Ambry Genetics to NM_182918.4(ERG):c.1088A>G (p.Asp363Gly), citing Ambry Variant Classification Scheme 2023: The c.1109A>G (p.D370G) alteration is located in exon 12 (coding exon 10) of the ERG gene. This alteration results from a A to G substitution at nucleotide position 1109, causing the aspartic acid (D) at amino acid position 370 to be replaced by a glycine (G). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
