NM_000245.4(MET):c.1625A>G (p.His542Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H542R variant (also known as c.1625A>G), located in coding exon 4 of the MET gene, results from an A to G substitution at nucleotide position 1625. The histidine at codon 542 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
