Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014727.3(KMT2B):c.6976A>G (p.Met2326Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 2326 of the KMT2B protein (p.Met2326Val). This variant is present in population databases (no rsID available, gnomAD 0.001%). This variant has not been reported in the literature in individuals affected with KMT2B-related conditions. ClinVar contains an entry for this variant (Variation ID: 2579594). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on KMT2B protein function. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
