Pathogenic for Duchenne muscular dystrophy — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to GRCh38/hg38 Xp21.1(chrX:32536377-32771300)x0, citing ACMG/ClinGen CNV Guidelines, 2019: A hemizygous deletion of exons 8-17 in DMD (NM_004006.3) was identified by exome sequencing and confirmed by genome sequencing in one male with Duchenne muscular dystrophy ([GRCh 38] chrX:32536377_32771300x0)(PMID: 32528171). Inheritance information is unavailable. The patient phenotype is nonspecific, but is consistent with cases described in the literature and/or published databases with overlapping variants. This exon deletion has also been reported in the literature in at least 2 hemizygous individuals with Duchenne muscular dystrophy (PMID: 31661024, 14977063). This intragenic variant is predicted to cause a frameshift, which alters the protein’s amino acid sequence beginning at exon 8 and leads to a premature termination codon. This alteration is then predicted to lead to a truncated or absent protein. Loss of function of DMD is an established disease mechanism in X-linked Duchenne muscular dystrophy (https://search.clinicalgenome.org/kb/gene-dosage). In summary, this variant meets criteria to be classified as pathogenic for X-linked Duchenne muscular dystrophy.
