This variant lies in the SAMD9L gene (transcript NM_152703.5) at coding-DNA position 2734, where C is replaced by G; at the protein level this means replaces glutamine at residue 912 with glutamic acid — a missense variant. Submitter rationale: The SAMD9L c.2734C>G (p.Gln912Glu) missense change is absent in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/). The in silico tool REVEL predicts a benign effect on protein function, but to our knowledge this prediction has not been confirmed by functional studies. In silico predictions have not been found to correlate with syndromic risk and are not considered supporting evidence of a pathogenic or benign effect (PMID: 34621053). To our knowledge, this variant has not been reported in individuals with SAMD9L-related disease.
