Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.1911C>G (p.Phe637Leu), citing Ambry Variant Classification Scheme 2023: The p.F637L variant (also known as c.1911C>G), located in coding exon 15 of the POLD1 gene, results from a C to G substitution at nucleotide position 1911. The phenylalanine at codon 637 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
