Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000071.3(CBS):c.1346T>G (p.Val449Gly), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CBS c.1346T>G (p.Val449Gly) results in a non-conservative amino acid change located in the CBS domain (IPR000644) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 165690 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1346T>G in individuals affected with Homocystinuria has been reported. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no change in enzyme activity (Mendes_2014). The following publication have been ascertained in the context of this evaluation (PMID: 25044645). No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
