This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 4058, where G is replaced by C; at the protein level this means replaces tryptophan at residue 1353 with serine — a missense variant. Submitter rationale: This missense variant replaces tryptophan with serine at codon 1353 of the ATP7B protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). Although functional studies have not been reported for this variant, it alters a conserved tryptophan residue in the transmembrane M6 domain of the ATP7B protein (a.a. 1352 - 1375), a highly conserved region that is considered to be important for ATP7B protein function (PMID: 35245129; ClinVar). This variant has been reported in one individual affected with Wilson disease (PMID: 30230192). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
