This variant lies in the ARSB gene (transcript NM_000046.5) at coding-DNA position 160, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 54 with histidine — a missense variant. Submitter rationale: Variant summary: ARSB c.160G>C (p.Asp54His) results in a non-conservative amino acid change located in the sulfatase N terminal domain (IPR000917) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 175716 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.160G>C in individuals affected with Mucopolysaccharidosis Type VI (Maroteaux-Lamy Syndrome) and no experimental evidence demonstrating its impact on protein function have been reported.
