The exact breakpoint at the 5' end of this variant is unknown and therefore this duplication might extend upsream of the assayed region of the GNB4 gene. A presumed nomenclature of c.(?_-281)_(96+1_97-1)dup has been designated for the purposes of this classification. It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). Since the exact breakpoints of this duplication are not known, it is not possible to predict if it causes an in-frame or an out-of-frame product. The variant was absent in 21694 control chromosomes (gnomAD, structural variants dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(?_-281)_(96+1_97-1)dup in individuals affected with Charcot-Marie Disease Dominant Intermediate F and no experimental evidence demonstrating its impact on protein function have been reported.
