Submitter rationale: Variant summary: CHST6 c.815G>A (p.Arg272His) results in a non-conservative amino acid change located in the Sulfotransferase domain (IPR000863) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4.1e-06 in 245608 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.815G>A has been reported in the literature in at least one compound heterozygous individual affected with Macular Corneal Dystrophy with a second variant of unknown pathogenicity (e.g. Klintworth_2006). These report(s) do not provide unequivocal conclusions about association of the variant with Macular Corneal Dystrophy.
