This variant lies in the MPL gene (transcript NM_005373.3) at coding-DNA position 398, where C is replaced by T; at the protein level this means replaces proline at residue 133 with leucine — a missense variant. Submitter rationale: Variant summary: MPL c.398C>T (p.Pro133Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 250492 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.398C>T has been reported in the literature in a family affected with Thrombocytosis without evidence of causality (Dasouki_2021). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
