NM_024426.6(WT1):c.1390A>G (p.Lys464Glu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K459E variant (also known as c.1375A>G), located in coding exon 9 of the WT1 gene, results from an A to G substitution at nucleotide position 1375. The lysine at codon 459 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
