Submitter rationale: This DLL4 missense variant (rs759324100) is absent from a large population dataset and has not been reported in ClinVar nor the literature, to our knowledge. Two bioinformatic tools queried predict that this substitution would be tolerated. The valine residue at this position is evolutionarily conserved across most mammalian species assessed; however a leucine residue is present at this position in three species. We consider the clinical significance of DLL4 c.1429G>T to be uncertain at this time.
