NM_000153.4(GALC):c.911C>T (p.Thr304Ile) was classified as Pathogenic for Galactosylceramide beta-galactosidase deficiency by Department of Medical Genetics, National Institute of Health, citing ACMG Guidelines, 2015: The c.911C>T(p.Thr304Ile) biallelic variant in GALC gene was detected via Next generation sequencing in a male Moroccan patient with Krabbe disease. It is classified as Variant with Uncertain Significance according to the ACMG guidelines. The presence of this variant was confirmed via Sanger sequecing in the patient. It has not been previously reported and was not found in 138 Moroccan Clinical Exomes on our house database. Therefore, this variant is associated with Krabbe disease in our patient and meets our criteria to be classified as pathogenic.
