NM_000492.4(CFTR):c.1709T>A (p.Leu570Ter) was classified as Pathogenic for Cystic fibrosis by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFTR c.1709T>A (p.Leu570X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 250474 control chromosomes in GnomAD. c.1709T>A has been reported in a compound heterozygous state with a common pathogenic variant F508del (DV) in an individual affected with Cystic Fibrosis (Dayanga-Erden_2020). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 32687833). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as pathogenic.
