Submitter rationale: Variant summary: ACADM c.905A>G (p.Tyr302Cys) results in a non-conservative amino acid change located in the Acyl-CoA dehydrogenase/oxidase C-terminal (IPR009075) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250862 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.905A>G has been reported in the literature in compund heterozygous individuals affected with Medium Chain Acyl-CoA Dehydrogenase Deficiency without evidence of cosegregation provided (Gong_2021, Li_2022). These reports do not provide unequivocal conclusions about association of the variant with Medium Chain Acyl-CoA Dehydrogenase Deficiency. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33841490, 36068006).
