This variant lies in the RHAG gene (transcript NM_000324.3) at coding-DNA position 827, where C is replaced by G; at the protein level this means replaces threonine at residue 276 with serine — a missense variant. Submitter rationale: Variant summary: RHAG c.827C>G (p.Thr276Ser) results in a conservative amino acid change located in the Ammonium transporter AmtB-like domain (IPR024041) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251402 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.827C>G in individuals affected with Rh-Null, Regulator Type and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
