Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000041.4(APOE):c.737G>C (p.Arg246Pro), citing Ambry Variant Classification Scheme 2023: The p.R246P variant (also known as c.737G>C), located in coding exon 3 of the APOE gene, results from a G to C substitution at nucleotide position 737. The arginine at codon 246 is replaced by proline, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
