NM_000051.4(ATM):c.2304T>G (p.Asn768Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N768K variant (also known as c.2304T>G), located in coding exon 14 of the ATM gene, results from a T to G substitution at nucleotide position 2304. The asparagine at codon 768 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
