Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.3014A>G (p.Lys1005Arg), citing Ambry Variant Classification Scheme 2023: The p.K1005R variant (also known as c.3014A>G), located in coding exon 16 of the NPAT gene, results from an A to G substitution at nucleotide position 3014. The lysine at codon 1005 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
