Uncertain significance — the classification assigned by Ambry Genetics to NM_005896.4(IDH1):c.661G>C (p.Gly221Arg), citing Ambry Variant Classification Scheme 2023: The p.G221R variant (also known as c.661G>C), located in coding exon 4 of the IDH1 gene, results from a G to C substitution at nucleotide position 661. The glycine at codon 221 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
