Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_198578.4(LRRK2):c.4514T>A (p.Ile1505Lys), citing Ambry Variant Classification Scheme 2023: The p.I1505K variant (also known as c.4514T>A), located in coding exon 31 of the LRRK2 gene, results from a T to A substitution at nucleotide position 4514. The isoleucine at codon 1505 is replaced by lysine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
