This variant lies in the SMARCA4 gene (transcript NM_001387283.1) at coding-DNA position 4248, where C is replaced by G; at the protein level this means replaces phenylalanine at residue 1416 with leucine — a missense variant. Submitter rationale: The p.F1416L variant (also known as c.4248C>G), located in coding exon 29 of the SMARCA4 gene, results from a C to G substitution at nucleotide position 4248. The phenylalanine at codon 1416 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Missense and in-frame variants in SMARCA4 are known to cause neurodevelopmental disorders; however, such associations with rhabdoid tumor predisposition syndrome including small cell carcinoma of the ovary-hypercalcemic type (SCCOHT) are exceedingly rare (Kosho T et al.
