Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.104A>T (p.Lys35Met), citing Ambry Variant Classification Scheme 2023: The p.K35M variant (also known as c.104A>T), located in coding exon 1 of the CDK4 gene, results from an A to T substitution at nucleotide position 104. The lysine at codon 35 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
