Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002354.3(EPCAM):c.671C>G (p.Ser224Cys), citing Ambry Variant Classification Scheme 2023: The p.S224C variant (also known as c.671C>G), located in coding exon 7 of the EPCAM gene, results from a C to G substitution at nucleotide position 671. The serine at codon 224 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
