Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1536G>T (p.Glu512Asp), citing Ambry Variant Classification Scheme 2023: The p.E512D variant (also known as c.1536G>T), located in coding exon 9 of the DICER1 gene, results from a G to T substitution at nucleotide position 1536. The glutamic acid at codon 512 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
