Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4345C>A (p.His1449Asn), citing Ambry Variant Classification Scheme 2023: The p.H1449N variant (also known as c.4345C>A), located in coding exon 22 of the DICER1 gene, results from a C to A substitution at nucleotide position 4345. The histidine at codon 1449 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
